肾综合征出血热疫苗株Z10和汉坦病毒西安分离株XAAa10091712亲缘关系以及免疫效果研究

摘要 目的研究目前使用的双价肾综合征出血热疫苗构成株Z10与汉坦病毒西安分离株XAAa10091712的相互关系,为西安地区肾综合征出血热防治提供依据。方法通过比对疫苗构成株Z10和西安分离株核苷酸和氨基酸序列差异以及疫苗注射后产生抗体对西安分离株中和作用研究两者相互关系。结果基因组序列比对显示两毒株S,M和L片段核苷酸相似性为89.5%,87.8%和87.8%,核蛋白、糖蛋白和RNA聚合酶相似性分剐为98.4%,93.2%和96.3%;疫苗接种后产生针对N蛋白IgG类抗体在免疫前、基础免疫后、加强后分别为25.50%,62.54%和89.06%;针对N蛋白IgG抗体阳性标本中中和抗体阳性率为20.0%。结论疫苗株与西安分离株基因组序列相似性均在90%以下,糖蛋白氨基酸序列差异为6.8%;疫苗接种后针对西安分离株产生中和抗体阳性率较低。 Objective To study the relationship between the vaccine strain Z10 of Hemorrhagic fever with renal syndrome and viral isolate XAAa10091712. Methods Compared the sequences difference of nucleotide and amino acid of vaccine strain Z10 and viral isolate XAAa10091712. Detected the neutralization antibody against viral isolate after vaccination. Results The similarity of nucleotide sequence of S, M and L between two strains was 89.5 %, 87.8 % and 87.8 % respectively, the similarity of the matching amino acid sequence was 98.4 %, 93.2 % and 96.3 % respectively. The positive rate of IgG antibody against hantavirus N protein in prevaccination, after basic vaccination, after enhanced vaccination were 25.50 %,62.54 % and 89. 06% respectively. The neutralization antibody positive rate in IgG positive samples was 20. 0%. Conclusion The vaccine strain and viral isolate in Xi' an were a bit different in nucleotide sequence and the difference of glycoprotein reached 6.8 %, and the neutralization antibody positive rate was rather low.
