摘要
目的制备开管毛细管电色谱柱分离混合单糖。方法采用溶胶-凝胶法,选择β-环糊精(β-CD)作为固定相,制备开管毛细管电色谱柱,并用于1-苯基-3-甲基-5吡唑啉酮(PMP)衍生的混合单糖的分离。结果硼砂缓冲液浓度20 mmol.L-1、pH值5.5、电压15 kV、进样时间2 s、乙腈及二甲基亚砜(DMSO)的用量分别为5%时,混合单糖可得到良好分离。结论开管毛细管电色谱分离糖类物质与区带电泳相比,显著降低了缓冲液中非挥发性盐的浓度,可更好地实现与质谱联用。
Objective To prepare β-CD open tube capillary electrochromatography column to separate mixed mono- saccharide. Methods Sol-gel was used to prepare open tube capillary eletrochromatography column coated with β-cyclodextrin, and the 1-phenyl-3methyl-5-phrazolone derivative of the mixed monosaccharide was separated by the col- umn. Results The mixed monosaccharide was separated under the following conditions: buffer solution, 20 mmol .L - 1 borax-DMSO 5%, acetonitrile 5%, pH 5.5; detection wavelength, 245 nm; voltage, 15 kV; room temperature; injection for 2 s. Conclusion Compared with capillary zone electrophoresis, open tube capillary eletrochromatography may dramati- cally reduce the concentration of non-volatile salt in buffer solution in the separation of mixed monosaccharide.
