妊娠高血压综合征患者血浆对体外培养的内皮细胞合成血管活性物质的影响 被引量:3

The Effect of Plasma from Women with Preeclampsia on Endothelial Cells Activators in vitro

摘要 目的 探讨妊娠高血压综合征 (妊高征 )患者的血浆对体外培养的内皮细胞合成血管活性物质的影响 ,和妊高征患者血浆中是否存在某些引起内皮细胞损伤的物质。方法 应用放射免疫法测定 2 0例先兆子痫患者 (先兆子痫组 )及 15例正常晚期妊娠妇女 (正常晚孕组 )产前血浆前列环素(PGI2 )代谢产物 6 keto PGF1α、血栓素 (TXA2 )代谢产物TXB2 的水平。在体外培养的牛肺动脉内皮细胞中分别加入先兆子痫患者和正常晚期妊娠妇女的血浆 ,测定 2 4h后内皮细胞释放的 6 keto PGF1α和TXB2 水平。结果  (1)先兆子痫组血浆 6 keto PGF1α水平为 (94.49± 2 5 .2 3)ng/L ,TXB2 水平为(10 4.6 1± 13.12 )ng/L ,TXB2 / 6 keto PGF1α比值为 1.11± 0 .0 3。正常晚孕组血浆 6 keto PGF1α水平为(2 48.81± 5 1.99)ng/L ,TXB2 水平为 (6 6 .2 6± 38.80 )ng/L ,TXB2 / 6 keto PGF1α比值为 0 .2 8± 0 .0 2。与正常晚孕组比较 ,先兆子痫组血浆 6 keto PGF1α水平显著降低 (P <0 .0 1) ,TXB2 水平显著升高 (P<0 .0 1) ;两组的TXB2 / 6 keto PGF1α比值比较 ,差异有极显著性 (P <0 .0 1)。 (2 )加入先兆子痫患者血浆的内皮细胞释放的 6 keto PGF1α为 (136 3.0 0± 99.16 )ng/L ,加入正常晚期妊娠妇女血浆的内皮? Objective To study the effect of plasma from women with preeclampsia on endothelial cell activators in vitro and determine whether factor (s) in the plasma of women with preeclampsia induces activation of vascular endothelial cells. Methods Twenty women with preeclampsia and 15 normal ones at late trimester of pregnancy were studied, from whom maternal venous blood samples were collected. Plasma 6 keto PGF1α, the end products of prostacyclin (PGI 2) and TXB 2, the end products of thromboxane A 2 (TXA 2), were measured by radioimmunoassay. Plasma from women with preeclampsia or from normal women at late trimester was added to the bovine pulmonary microvessel endothelial cells in vitro. Twenty four hours later, levels of 6 keto PGF1α produced by cultured endothelial cells were measured. Results The mean values of 6 keto PGF1α in matrnal venous plasma were (94.49±25.23) ng/L in preeclamptic group and (248.81±51.99) ng/L in normal pregnant group. Plasma TXB 2 values were (104.61±13.12) ng/L in preeclamptic group and (66.26±38.80) ng/L in normal pregnant group. TXB 2/6 keto PGF1α ratios were (1.11±0.03) in preeclamptic group and (0.28±0.02) in normal pregnant group. Maternal plasma values of 6 keto PGF1α were lower in peeclamptic group than those of normal pregnant group ( P <0.01). On the other hand, plasma TXB 2 values were higher in preeclamptic group than in normal pregnant group ( P <0.01). TXB 2/6 keto PGF1α ratios, however, were significantly different between the two groups ( P <0.01). The values of 6 keto PGF1α produced by cultrued endothelial cells exposed to 2% plasma were (1 363.00±99.16) ng/L in preeclamptic group and (819.49±96.62) ng/L in normal pregnant group ( P <0.01). Conclusions PGI 2 and TXA 2 may play an important role in preeclampsia. Plasma from women with preeclampsia could stimulate the production of prostacyclin in cultured endothelial cells in vitro.
