摘要
非类固醇消炎止痛药 (NSAIDs)已被广泛用于治疗炎症发热、止痛及抗血小板活性。近期 ,临床、流行病学研究及动物实验均有证据表明NSAIDs尚有防癌作用 ,有可能成为临床预防癌症的方便途径。其可能的机制为 :1、抑制环氧化酶 1、2。 2、影响细胞周期 ,包括对增生与分化、肿瘤抑制基因或促进基因的影响及对细胞周期的抑制。 3、免疫监视。 4、增加酰基鞘氨醇的生成。 5、抑制细胞核κ因子 (NFκB)。然而 ,确切机制仍不十分清楚 。
Non steroidal Anti inflammatory Drugs(NSAIDS) have been extensive used for treatment of inflammation,fever as agents of analgesia and anti platelet activity. Recently, lots of clinical,epidemic study and animal experimental evidences have shown that NSAIDs may be as agents for cancer chemoprevention. The possible mechanisms are (1) inhibition of cyclo oxytenase 1 and 2(2) alteration of cell cycle,including alterations in proliferation and differentiation alterations of tumor suppressor/promoter genes, and arrest of cellcyle,(3) immune surveillamce,(4) increased production of ceramide,and (5) inhibition on nuclear factor kappa B(NF κB). However, the exact mechanism of the action remains unclear and need further research.
