摘要
目的观察携带人尾型同源盒转录因子2(Cdx2)基因沉默的重组慢病毒颗粒(Cdx2-RNAi-LV)转染人胃癌MGC-803细胞对中药人参黄芪复方药物敏感性的影响。方法采用半定量逆转录-聚合酶链反应(RT-PCR)和Western blot检测Cdx2基因表达;MTT方法检测细胞的增殖抑制率;Giemsa染色检测细胞的克隆形成;流式细胞仪检测细胞的凋亡;细胞划痕实验检测细胞迁移能力。结果含Cdx2基因沉默的重组慢病毒颗粒转染人胃癌MGC-803细胞有效沉默Cdx2基因表达;中药人参黄芪复方能抑制人胃癌MGC-803细胞生长、抑制细胞克隆形成、诱导细胞凋亡、抑制细胞迁移;Cdx2基因沉默转染胃癌MGC-803细胞组应用中药人参黄芪复方,其细胞增殖抑制率、细胞凋亡率明显高于单用中药未转染组(P<0.05),其细胞克隆形成、细胞迁移面积均显著低于单用中药未转染组(P<0.05)。结论 Cdx2基因沉默转染人胃癌MGC-803细胞,可明显提高胃癌细胞对中药人参黄芪复方的敏感性。
Objective To investigate the effect of transfected Cdx2 gene silencing of recombinant lentivirus particles(Cdx2-RNAi-LV) on drug sensitivity of traditional Chinese medicine(TCM) compound of Ginseng and Astragalus in human gastric cancer MGC-803 cells.Methods The expression of Cdx2 gene was assessed by reverse transcription polymerase chain reaction(RT-PCR) and Western blotting;the cell proliferation was assessed by MTT assay;the cell apoptosis was observed by flow cytometry;the cell colony of formation was detected by Giemsa staining;and the cell migration was assessed by cell scratch experiment.Results Cdx2 gene was successfully silenced by transfected Cdx2 gene silencing recombinant lentivirus particles in human gastric cancer MGC-803 cells.The TCM compound of Ginseng and Astragalus could inhibit the cellular growth,inhibit the colony formation,induce cell apoptosis,reduce cell migration of the human gastric cancer cell line MGC-803l;which was significantly promoted by Cdx2 silencing when comparing with those using TCM compound alone(P<0.05).Conclusion Cdx2 gene silencing increases the the drug sensitivity of TCM compound of Ginseng and Astragalus in gastric cancer cells.
