摘要
【目的】采用近红外漫反射光谱法对制川乌中的相关成分进行快速定量分析。【方法】应用近红外漫反射光谱法采集119个制川乌样品的近红外光谱数据,采用偏最小二乘回归法分别建立近红外光谱信息与单酯型生物碱类成分、双酯型生物碱类成分、水分之间的定量分析模型。【结果】制川乌中单酯型生物碱类成分、双酯型生物碱类成分、水分校正模型的建模光谱范围为8 431.26~4 075.93,5 086.76~4 088.64、7 725.44~5 924.25,7 934.87~4 088.64 cm-1,校正模型的校正均方根误差(RMSEC)单酯型生物碱类成分=0.098,相关系数(r)单酯型生物碱类成分=0.955 9;RMSEC双酯型生物碱类成分=0.034,r双酯型生物碱类成分=0.969 4;RMSEC水分=0.002,r水分=0.976 7,验证模型的预测均方根误差(RMSEP)单酯型生物碱类成分=0.168,r单酯型生物碱类成分=0.926 6;RMSEP双酯型生物碱类成分=0.039,r双酯型生物碱类成分=0.982 6;RMSEP水分=0.004,r水分=0.836 3。3类成分的预测值与参考值的相关系数分别为0.970、0.993、0.918(均P<0.001,说明近红外光谱法与高效液相色谱法测定生物碱类成分相关性较好,近红外光谱法与烘干法测定水分相关性较好,可以较准确地预测其覆盖范围的相应物质含量。【结论】近红外光谱法具有不破坏样品、不需要对样品进行复杂的前处理、分析速度快、预测结果准确等特点,可用于制川乌中相应成分含量的预测。
Objective To make a rapid analysis of the constituents of Radix Aconiti Preparata (RAP) by near- infrared diffuse reflectance spectroscopy (NIDRS). Methods The NIDRS data of 119 RAP samples was collected for the establishment of quantitative analysis models of monoester alkaloids, diester alkaloids and water content NIDRS information by partial least squares (PLS) regression method. Results The modeling spectral ranges of RAP monoester alkaloids, diester alkaloids, and water content were 8 431.26-4 075.93, 5 086.76-4 088.64, 7 725.44-5 924.25, and 7 934.87-4 088.64 cm-1 respecitvely. The root mean square error of calibration (RMSEC) for monoester alkaloids, diester alkaloids, and water content was 0.098, 0.034, and 0.002, and the correlation coefficient (r) of their calibration models was 0.955 9, 0.969 4, and 0.976 7, respectively. The root mean square error of prediction (RMSEP) for monoester alkaloids, diester alkaloids, and water content was 0.168, 0.039, and 0.004, and the correlation coefficient of their validation models was 0.926 6, 0.982 6 and 0.836 3, respectively. The correlation coefficient of the predicted value with the reference value of monoester alkaloids, diester alkaloids, and moisture was 0.970, 0.993 and 0.918, respectively (P〈0.001), indicating that NIDRS had the similar effect to high pressure liquid chromatography (HPLC) for the assay of alkaloids, a nd NIDRS was as good as oven drying method for the assay of water content. Conclusion NIDRS has the advantages of rapid analysis and accurately predicting results, without destroying the samples or complex pre-processing, which can be used for the prediction of corresponding ingredients of RAP.
