摘要
目的探讨抑癌基因PTPN12在肝癌细胞系Hep-G2中生物治疗肝癌的效果,揭示PTPN12在临床应用上的潜在意义。方法在Hep-G2细胞中顺转、稳定转染PTPN12基因后,通过MTT、WesternBlot、荧光定量PCR分析抗肿瘤作用。结果转染PTPN12后的Hep-G2细胞增殖慢,凋亡率高,PTPN12蛋白表达量高。结论 PTPN12基因在肝癌表达上调,能有效抑制肿瘤细胞的生长,从而促进肿瘤细胞的凋亡。因此,以PTPN12为靶点开展生物治疗可能是一种新的治疗手段。
Objective To investigate the anti-tumor effect of PTPN12 in Hep-G2 cells, and to reveals the hidden meaning of PTPN12 in the clinical application. Methods The PTPN12 gene was transfected into Hep-G2 cells, and then the anti-tumor effect was analyzed by Real-time RT-PCR, Western Blot, and MTT. Results Increasing PTPNI2 expression in Hep-G2 cells were observed. The proliferation of Hep-G2 cells was inhibited, and the apoptosis rate was high. Conclusion PTPN12 may be a novel strategy for bio-therapeutic regents in liver cancer, which can ef- fectively inhibit the growth of tumor cells and promote apoptosis.
