摘要
目的:建立α-乙酰乳酸脱羧酶中四环素残留量测定高效液相色谱法。方法:采用EDTA-Mcllvaine缓冲液提取样品,提取液经HLB固相萃取柱净化,以C18柱分离,乙腈-甲醇-0.01mol/L草酸溶液(12∶6∶82)为流动相,紫外检测器检测波长为350nm。结果:四环素的检测限(LOQ)为0.04mg/L;线性范围0.09mg/L^15.0mg/L;回收率为82.5%,RSD=2.6%(n=6)。结论:本实验建立的方法简便灵敏,结果准确可靠,重现性好,适用于α-乙酰乳酸脱羧酶中四环素残留量的测定。
Objective: To establish a HPLC method for the determination of tetracycline in et-Acetolactate decarboxylase. Method: Samples were extracted with EDTA-Mcllvaine buffer solution, and then, the extraction was purified by HLB solid phase extraction column. Chromatographic separation was carried out on a C18 column, the mobile phase is methanol-acetonitrile -0.01mol/L oxalic acid solution, detected by DAD (350nm). Results: Calibration curves were linear within the range of 0.09mg/L-1 5.0mg/L and the limit of detection were 0.04mg/L. The recovery were 82.5% and the RSD was 2.6%(n=6). Conclusion: This method is simple, sensitive, accurate for the determination of tetracycline in α-acetolactate decarboxylase.
