摘要
目的:探讨血管内皮生长因子(VEGF)对关节软骨细胞mmP-1及mmP-3表达的影响。方法:原代培养SD乳鼠的关节软骨细胞。实验分为4组,每组加入不同因素:A组(对照组)不加任何处理因素;B组:10ng/ml VEGF;C组:10ng/ml IL-1β;D组:10ng/mlVEGF+10ng/ml IL-1β,应用定量PCR方法检测软骨细胞中mmP-1及mmP-3mRNA的表达水平。结果:mmP-1 mRNA表达水平:与A组比较,B、C、D组明显升高,B、C组明显低于D组。mmP-3 mRNA表达水平:与A组(对照组)比较,B、C、D组mRNA表达明显升高。D组明显高于B、C组。结论:VEGF能够明显上调软骨细胞mmP-1、-3的mRNA表达。
Objective: To investigate the effect of VEGF on mmP-1 and mmP-3 expression in articular chondrocytes.Methods: Primary cultured SD rat articular chondrocytes.The experiment was divided into 4 groups: group A(control group) without any treatment;B group 10ng/ml VEGF and group C group 10ng/ml IL-1;group D group 10ng/ml VEGF+10ng/ml IL-1β.Quantitative PCR was used to observe the mmP-1/-3 mRNA and TIMP-1/TIMP-3 mRNA expression.Results: mmP-1 mRNA expression level: B,C and D was significantly higher than that in group A(control group),and in groups D it was significantly higher than that in group B and C.mmP3 mRNA expression level: B,C and D was significantly higher than that in group A(control group),and in groups D it was significantly higher than that in group B and C.Conclusion: VEGF can increase the mmP-1,mmP-3 mRNA expression in the chondrocytes.
