摘要
目的:观察波动性高血糖对糖尿病大鼠骨质疏松的影响。方法:将SD大鼠随机分为3组,分别为:正常对照组(N)、糖尿病稳定性高血糖组(D)和糖尿病波动性高血糖组(F)。SD大鼠腹腔注射链脲佐菌素(STZ)诱导建立糖尿病模型,F组每日定时腹腔注射普通胰岛素,并错时给予葡萄糖,造成一日中血糖浓度大幅度波动模型。一段时间后,观察大鼠骨密度、骨代谢等变化。结果:血清生化指标显示,相对于持续高血糖,波动性高血糖促进骨吸收,抑制骨生成的作用更强。而血糖波动的水平与与胰岛素样生长因子-1、胰岛素、胰岛素抵抗指数呈正相关,与HbA1c水平无关。骨组织学切片显示,糖尿病可引起大鼠骨质疏松,而波动性高血糖相对于持续高血糖,能显著加重骨质疏松的程度。结论:波动性高血糖对大鼠骨质疏松的影响重于稳定性高血糖。
Objective: To observe the effects of fluctuant high blood glucose on osteoporosis. Methods: SD rats were randomly divided into 3 groups : normal control group (N) , stable high blood glucose group (D) and fluctuant high blood glucose group (F). Diabetic rats were induced by intraperitoneal injection of STZ. The fluctu- ant high blood glucose animal model was induced by intraperitoneal injection of ordinary insulin and glucose at dif- ferent time point every day. After a period of time change of bone mineral density and bone metabolism were ob- served. Result: Serological indicators showed that compared with stable high blood glucose, the fluctuant high blood glucose had stronger effect on stimulates bone absorption and inhibition of bone formation. The fluctuantions levels of blood glucose was positively related with levels of IGF-1, INS, IR, whereas not absolute correlation with HbA1 c levels. Bone histological sections showed diabetes can cause osteoporosis. Compared with stable high blood glucose, fluctuant high blood glucose significantly increased the degree of osteoporosis. Conclusion: Fluetuant high blood glucose lead to more sever osteoporosis than stable high blood glucose.
