摘要 缺氧诱导因子-1(hypoxia-inducible factor-1,HIF-1)是在缺氧时诱导产生的一类核转录因子。大量研究证实HIF-1的亚单位HIF-1α在许多肿瘤中过量表达,与肿瘤的生长、侵袭、转移及患者的预后等密切相关,而降低HIF-1α活性对肿瘤的生长有明显抑制作用。因此,HIF-1α在临床上可作为肿瘤的诊断及判断预后的指标,以HIF-1α为靶点的治疗有望成为肿瘤治疗的新途径。近年来有关HIF-1α与胃肠道间质瘤(Gastrointestinal Stromal Tumors,GIST)关系的研究有所进展。本文将对HIF-1α在肿瘤中的作用机制及其与GIST关系的研究进展作一综述。 Hypoxia-inducible factor-1(HIF-1) is a nuclear transcription factor induced in the hypoxia environment. A number of studies have demonstrated that one of HIF-1 subunits(HIF-1 α) is over- expressed in many tumors, and it is closely related to tumor growth,invasion, metastasis and the prognosis of the patients. And reduce the activity of HIF-1 α has significant inhibitory effects on tumor growth. Therefore, HIF-1 α can be used as the index to diagnosis and prognosis of the tumor in clinical, Using HIF-1 α as a therapeutic target is expected to become a new way to treat cancer. In recent years, research about the relationship of HIF-1 α and gastrointestinal stromal tumors(GIST) is progressed. This review will summarizes the research progress on the mechanism of action of HIF-1 in tumors and its relationship with GIST.
