籼稻体细胞无性系雄性不育突变体后代遗传及其恢保关系鉴定

Inheritance and identification of restoring and maintaining relationship of somaclonal malesterile mutants in indica rice

摘要 对野败型保持系珍汕 97B、恢复系 IR2 4、IR2 6和泰引 1号等 4个水稻材料的幼穗在不同的培养基上培养、再生植株及对其后代进行育性和测交鉴定及其它遗传分析 ,探讨了雄性不育变异株的恢保关系 ,结果表明 :在 IR2 6、泰引 1号和珍汕 97B的 R1 、 R2 群体中都获得了不同比率的雄性不育变异株 ,这些不育株的花粉败育可分为无花粉、典败、圆败和染败 4种类型。对离体培养产生的雄性不育变异株用一批现有 CMS不育系的保持系和恢复系进行杂交 ,其中有 2株分别来自 IR2 6(编号为 2 691 57)和珍汕 97B (编号为 B2 91 54)的不育株 ,其恢保关系与野败型一致 ,现已回交 5代。来自 IR2 6(编号为 2 692 4 1 1 )的另一雄性不育株 ,用上述亲本已连续回交 5代 ,后代均不育 ,其恢保关系似与野败不同。而有些来自珍汕 97B和 IR2 6的 1 2株雄性不育变异株 ,杂交后代( F1 )全部可育 ,结实率 Young panicles from four rice varieties,Zhenshan 97B(WA type maintainer line indica ),IR24,IR26,Taiyin 1(WA type restorer line, indica ),were cultured on different media.Regenerated palnts were obtained.Pollen fertility was examined.Testcrosses of malesterile variants were made.Genetic analyses of malesterile variants were done.The restoring and maintaining relationship of malessterile variants were discussed.The results showed that 15 cases of malesterilevariants in three rice varieties,Zhenshan 97B,IR26 and Taiyin 1 were found,in which 10 cases belonged to R 1 generation,and 5 to R 2 generation.The malesterile variation frequency in R 1 generation was 1 04%(1 01%~1 07%).One chimera was obtained from R 1 regenerated plants of IR26.Fertility segregation was found from R 2 generation of Taiyin 1 and Zhenshan 97B,respectively and the average malesterile variation frequency in Zhenshan 97B and Taiyin 1 was 2 22% and 1 89%,respectively.Abortive pollen of rice could be classified into four types;pollen free type (PFT),typical abortion type (TAT),spherical abortion type (SpAT) and stained abortion type(StAT). Malesterile variants in R 1 and R 2 generations were crossed with ten typical maintainer lines and restorer lines (Zhenshan 97B,Erjiuai 4B,Hongyuan B,Baoyuan B,Taiyin 1,Minghui 63,Ce 64,IR24,IR26 and IR36)to investigate the restoring and maintaining relationship of these malesterile variants.The main results were as follows: There were two typical abortion type(TAT) variants in R 1 regenerated plants which derived from IR26(coded 269157) and Zhenshan 97B(coded B29154),and their restoring and maintaining relationship was the same as that of WA type.These variants had been backcrossed with their maintainer lines for five generations,and kept sterile all along. One malesterile variant,which derived from IR26(coded 2692411,TAT),kept sterile continuously for five generations when they were backcrossed with the ten parents stated above. The other twelve malesterile variants,which derived from Zhenshan 97B,IR26 and Taiyin 1,turned into fertility with a lot of fertile pollens and very hight self seed set rate(more than 80%) when they were crossed with the ten parents stated above.
