摘要
利用PCR-RFLP和Real-time PCR方法分别检测了H-FABP基因在玉山黑猪中的基因多态性分布和各组织中的表达情况,并对H-FABP基因在玉山黑猪和大白猪组织中的表达进行了差异性研究。结果表明,玉山黑猪H-FABP基因在HinfI位点表现多态性,在HaeIII位点为单一等位基因。同时,该基因主要在玉山黑猪心、肾、脂肪和骨骼肌组织中表达,且在心脏组织中的表达量最高。差异表达分析发现,玉山黑猪心和骨骼肌组织中H-FABP基因的表达均显著高于大白猪(P<0.05),差异倍数分别为3.34和3.42。
The H - FABP polymorphism and mRNA expression in different tissues of Yushanhei pigs were detected by PCR - RFLP and Real - time PCR. And the expression differences in Yorkshire and Yushanbei pigs were also studied. The results indicated that H - FABP gene of Yushanhei pigs had polymorphism at Hinf I - RFLP and all D allele at HaeIII - RFLP. In Yushanhei pigs, the H - FABPgene was mainly expressed in heart, kidney, fat and skeletal muscles, and had the highest expression in heart. The differential expression analysis showed that the expression level in heart and skeletal muscle were all significantly higher in Yushanhei pigs than in Yorkshire （ P 〈 0.05 ）, the difference multiples were 3.34 and 3.42.
