我国猪瘟病毒流行毒株的抗原性分析 被引量:3

Antigenic analysis of classical swine fever viruses in China by monoclonal antibodies

摘要 为了解我国猪瘟病毒流行毒株之间的抗原性差异,分离了来自我国不同年份、不同省份的22株猪瘟病毒流行毒株,采用间接免疫荧光方法,用28株抗瘟病毒单克隆抗体对这些流行毒株进行了抗原性分析。结果显示,22株猪瘟病毒流行毒株只与抗猪瘟病毒的单抗反应,与抗瘟病毒属其他成员的单抗无交叉反应。且毒株不同,与单抗谱的反应也不同,根据反应荧光的强弱可以分为3类,28株单抗的识别谱有所差异,其中单抗302,303可以识别所有的毒株,抗猪瘟病毒E2蛋白的单抗识别的毒株数多于抗Erns蛋白的单抗,不同基因型以及同一基因型的不同毒株与单抗谱的反应均呈现多样性,同一基因型不同年代的毒株与单抗谱的反应也有所不同。证实我国猪瘟病毒流行毒株之间存在抗原性差异。 To study antigenic differences of classical swine fever virus(CSFV) in China,22 field strains of CSFV were isolated from 13 provinces in China, grown in cell culture and characterized by indirect immunofluorescence assay with a panel of 28 monoclonal antibodies(McAbs) against pestiviruses. All 22 CSFV strains reacted only with CSFV-specific McAbs, not with those raised against bovine viral diarrhea virus I(BVDV1),BVDV2 and border disease virus. Those McAbs directed against CSFV E2 protein recognized more isolates than Erns and NS2/3. E2-specific McAbs, McAbs 303 and 302, reacted with all 22 strains, confirming their value in differentiating CSFV from other pestiviruses. Furthermore,all strains had different patterns of reactivity with CSFV based on isolated year and genetic type,indicating that antigenic differences are present among Chinese CSFV strains.
