摘要
目的通过体外经皮渗透试验评价跌打喷雾剂制备工艺的可行性和合理性。方法采用改良Franz扩散池,高效液相色谱法检测水杨酸甲酯和丁香酚的累积透过量,对实验动物皮肤及皮肤的用药部位、接收液、制剂的不同载药量等进行考察,建立离体经皮渗透试验的方法。结果以大鼠腹部皮肤为用药部位、50%乙醇/生理盐水为接收液、载药量8%跌打喷雾剂的体外经皮渗透试验效果较好。水杨酸甲酯和丁香酚的体外经皮渗透行为均可较好的拟合零级动力学方程。结论体外经皮渗透试验可以作为评价跌打喷雾剂制备工艺的可行性和合理性的方法,且该方法简便、可靠。
Objective Thread study transdermal penetration in vitro to estimate the feasibility and rationality on molding technics of Dieda spray. Methods Using HPLC to determine the cumulative percent releasing amount of methyl salicylate and eugenol, establishing the experimental method of transdermal penetration to review the experimental conditions of penetration such as the selection of animal skin, receptor solutions, and different drug loading rate of Dieda spray with improved Franz-pool. Results The abdomen skin of rat was selected as experimental skin. 50%ethanol/normal saline as the recaptor solution.The best drug loading was 8% Dieda spray. methyl salicylate and eugenol of in vitro percutaneous permeability behavior all can better fitting level zero dynamics equation.Conclusion The transdermal penetration in vitro is a good method to estimate the feasibility and rationality on molding technics of Dieda spray. The method is simple and reliable.
