人极低密度脂蛋白受体基因CGG重复序列多态性的检测 被引量:1

Detection of CGG repeat polymorphism in the human VLDL-R gene

摘要 极低密度脂蛋向受体(VLDL-R)基因5'非编码区起始密码子上游19bp处存在CGG三核苷酸重复序列。文献报道,一股正常人中该基因CGG的重复次数为 5~11次,呈多态性分布,且有民族差异。目前尚无我国人群的资料。为了解中国汉族人群VLDL-R基因CGG重复序列的分布情况,采用聚合酶链反应(PCR)、变性聚丙烯酸胺凝胶电泳和银染等技术,对湖北地区 160例正常汉族人的该重复序列进行了分析,共检出重复次数分别为5、8、9及11的4种等位基因和5/5、5/8、8/8、819、9/9、5/9、8/11及5/11等八种基因型。VLDL-R基因CGG重复序列在中国汉族人及其他国家人群中的分布不尽相同。 The VLDL-R gene contains a polymorphic triplet (CGG) repeat located 19hp upstream of the initiation codon in the 5' uncoding region. According to literature, the repeating times of CGG triplet in the western population ranges from 5 to I l, with racial differences. There are no data concerning Chinese population. To determine the distribution pattern of the CGG triplet of VLDL-R gene in Chinese Han nationality, 160 normal subjects of Han nationality in Hubei area were analysed, using polymerase chain reaction (PCR), denaturing polyacrylamide gel electrophoresis and AgNO, staining techniques. 4 alleies were found, the CGG repeating times were 5、8、9 and 11, respectively. 8 genotypes were found, they were 5/5, 5/8, 8/ 8, 8/9, 9/9, 5/9, 8/11, 5/11. It has been found that the distribution pattern of repeating times of CGG triplets in Han nationality is different from that of other populations.
