摘要
使用近交系Wistar(RT1k)大鼠行异位全小肠移植。术后第 2天开始给予肠外营养至第 1 0天 ,对照组 (n =1 0 )行常规全胃肠道外营养 (TPN)支持 ,肝细胞生长因子 (HGF)组 (n =1 0 )行常规TPN支持的同时加用HGF ,观察移植肠形态学及吸收功能。HGF组移植肠绒毛高度、隐窝深度、粘膜厚度及绒毛表面积均明显大于对照组 ,肠上皮细胞超微结构基本保持完好 ,对照组则出现明显线粒体肿胀 ,嵴短小紊乱和微绒毛萎缩。HGF组小肠移植大鼠血浆1 5N 甘氨酸丰度在 1、2、3h均明显高于对照组。HGF能维护大鼠移植小肠粘膜形态 ,维持移植肠上皮细胞超微结构完整 ,并能改善移植肠对氨基酸的吸收能力。
To study the effect of hepatocyte growth factor(HGF) on the mucosal morphology and absorption affer rat intestine transplantation. Total small intestine transplantion was made with inbred Wistar(RT1k) rats heterotopically,either total parenteral nutrition(TPN group, n =10) or hepatocyte growth factor supplemented TPN(HGF group, n =10) was used in recipients from POD2 to POD10. The result showed that significant in increase of mucosal villous height,crypt depth,mucosal thickness and villous surface area were found in HGF TPN group compared with TPN group.Normal ultrastructure of the graft was maintained in HGF TPN group,atrophied microvilli and broken mitochondrial crista were observed in TPN group. The amount of recipient plasma 15 N Glycine was found to be greater at the 1st,2nd,3rd hour time points in HGF-TPN group than in TPN group. Conclusion Hepatocyte growth factor(HGF) can improve mucosal morphology,preserve enterocyte ultrastructure,enhance amino acid absorption of isograft after small intestine transplantation in rat.
