疲劳型亚健康肾阳虚证血浆代谢组学研究 被引量:19

Plasma Metabonomic Research on Kidney Yang Deficiency in Individuals with Sub-Health Fatigue

摘要 目的探讨疲劳型亚健康肾阳虚证代谢组学特征表达谱。方法收集5例疲劳型亚健康肾阳虚证患者作为试验组,6例同龄健康人作为正常对照组。采用1H-核磁共振(1H-NMR)技术,利用MestReNova5.3.1软件对两组1H-NMR谱进行相位矫正和基线矫正,采用OPLS-DA进行分类建立模型,结果以积分矩阵图和载荷矩阵图表示,分析疲劳型亚健康肾阳虚证血浆代谢组学谱。结果两组间代谢组学谱存在差异,31个内源性代谢产物组间比较差异有统计学意义(P<0.05),疲劳型亚健康肾阳虚证关键代谢产物为乳酸、氧化型谷氨酰胺、丙酮酸、醛固酮、还原型烟酰胺腺嘌呤二核苷酸磷酸/烟酰胺腺嘌呤二核苷酸(NADPH/NADH)、苯丙氨酸、2-羟丁酸、还原型谷胱甘肽和谷氨酰胺。结论疲劳型亚健康肾阳虚证存在多个特征性代谢产物,可能是疲劳型亚健康肾阳虚证发生发展的基础。 Objective To research the metabonomic characteristics of kidney yang deficiency in individuals with sub-health fatigue. Methods There were 11 subjects selected from Nanfang Hospital Health Management Center volunteers. Five subhealth fatigue patients with kidney yang deficiency pattern were enrolled as the experimental group and six healthy persons of the same age were chosen as the normal control group. Metabonomic finger printing was acquired and analyzed by using 1 H-NMR, MestReNova5.3.1 software. Results There was a significant difference between the experimental group and normal control group in 31 endogenous metabolites (P〈0.05). The key metabolites of sub-health fatigue patients with kidney yang deficiency were lactate, oxidized glutathione, pyruvate, aldosterone, nicotinamide adenine dinueleotide phosphate/nieotinamide adenine dinucleotide hydrate (NADPH/NADH), phenylalanine, 2-Hydroxybutyric acid, glutathione and L-glutamine. These metabolites were highly correlated with energy metabolism, oxidative stress and hypothalamie-pituitary-adrenal axis. Conclusion The sub-health fatigue patients with kidney yang deficiency pattern have many characteristic metabolites. These characteristic metabolites are related to various function changes and may induce sub-health fatigue with kidney yang deficiency pattern.
