摘要
目的通过体内外试验对8-O-乙酰哈巴苷防治Epstein-Barr病毒(EBV)感染的药效作用进行评价。方法采用实时荧光定量PCR(Real-time RT-PCR)技术观察8-O-乙酰哈巴苷对B95-8细胞内EBV衣壳抗原(VCA)的抑制作用;采用腹腔注射醋酸(HAc)所致小鼠腹腔毛细血管通透性增高模型检测8-O-乙酰哈巴苷对炎症早期的毛细血管通透性亢进和渗出的抑制作用。结果 8-O-乙酰哈巴苷大、中、小剂量组对B95-8细胞内EBV衣壳抗原均具有明显的抑制作用(P<0.01);8-O-乙酰哈巴苷大、中两个剂量组显著抑制醋酸引起的小鼠腹腔毛细血管通透性增高(P<0.01)。结论 8-O-乙酰哈巴苷对EBV感染具有明显的抑制作用。其不仅对EBV衣壳抗原具有明显的抑制作用,并且可以减轻炎症早期的毛细血管通透性亢进和渗出现象。
Objective To assess the pharmacodynamic effects of 8-O-acetylharpagide in the preven- tion and treatment of Epstein - Barr virus (EBV) by in vivo and in vitro trial. Methods The real - time fluo- rescent quantitative PCR ( Real - time RT - PCR) technology was used to observe the inhibition of 8-O-acetyl- harpagide on EBV viral capsid antigen(VCA) in B95 -8 cell. By the intraperitoneal injection of acetic acid, the model of the increased celiac capillary permeability was prepared in mice for the detection of the inhibi- tion of 8-O-acetylharpagide on the hyperfunction and effusion of capillary permeability at the early inflamma- tion. Results In the high - dose, middle - dose and low - dose groups, 8-O-acetylharpagide presented obvi- ous inhibition on EBV - VCA in B95 - 8 separately ( P 〈 0.01 ). In the high - dose and middle - dose groups, 8-O-acetylharpagide inhibited significantly the increased celiac capillary permeability in the mice in- duced by acetic acid( P 〈 O. 01 ). Conclusion 8-O-acetylharpagide presents the obvious inhibition on EBV infection. It not only apparently inhibits EBV - VCA, but also alleviates the hyperfunction and effusion of cap- illary permeability at the early inflammation.
