脱脂棉替代尼龙毛分离T细胞用于免疫学实验教学的探讨 被引量:3

摘要 探讨脱脂棉和尼龙毛分离纯化外周血T淋巴细胞的方法和特点,建立脱脂棉分离T细胞的方法。采用密度梯度离心法分离人外周血单个核细胞,分别以脱脂棉和尼龙毛分离出T细胞,使用流式细胞仪鉴定细胞纯度和回收率,台盼蓝检测细胞活力。脱脂棉分离T细胞的纯度、回收率和存活率虽然都略低于尼龙毛分离法,但两者比较均无显著性差异(p>0.05)。结果显示脱脂棉分离T细胞是一种简易、快速、经济、纯度较高和基本上不损伤细胞活性的方法,除了可以代替尼龙毛用于免疫学实验教学中,也可作为开放实验培养学生的创新能力。 Through evaluating the effect and characteristics of two methods for isolating T lymphocytes with cotton and nylon wool, we established a method of isolating T lymphocytes with cotton. Human peripheral blood mononuclear cells (PBMC) were prepared with Ficoll-Hypaque density gradient cen- trifugation, human peripheral T lymphocytes were separated with cotton and nylon wool isolation re- spectively. The purity, viability and cell proliferation of T lymphocytes were determined with flow cy- tometry, and trypan blue exclusion respectively. There was no significant difference between isolating T lymphocytes with cotton and nylon wool ( P 〉0. 05 ). The results suggest the conclusion that the isola- tion of T lymphocytes with cotton method is simpler, less time-consuming, better yield and with litter or no damage to the cells
