摘要
目的探讨点阵CO2激光和Nd:YAG激光(准长脉宽1064nm波长)照射对自然老化小鼠皮肤基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶组织抑制剂-1(TIMP-1)表达的影响,并进一步探讨其嫩肤机制。方法用点阵CO2激光和Nd:YAG激光照射小鼠背部脱毛皮肤每间隔10天1次,连续4次,分别在照射后2周、1个月及3个月取材,用免疫组化法检测MMP-1和TIMP-1的表达并观察其动态变化。结果点阵CO2激光和Nd:YAG激光照射区MMP-1和TIMP-1阳性表达,以2周时最明显:点阵CO2激光照射区MMP-1灰度值(115.14±5.23)与TIMP-1灰度值(104.01±3.15)均低于Nd:YAG激光照射区MMP-1灰度值(121.75±4.39)与TIMP-1灰度值(109.26±3.88),即点阵CO2激光照射区阳性表达较高,差异有统计学意义(P<0.05)。TIMP-1从总体水平上强于MMP-1的表达。结论点阵CO2激光和Nd:YAG激光照射促进MMP-1和TIMP-1表达增加,提示其直接参与了真皮重塑,点阵CO2激光组较Nd:YAG激光组效果明显。
Objective To detect the MMP-1 and TIMP-1 of the skins after application of fractional CO2 laser and Nd : YAG laser in aging mice, and primarily explore the mechanism of light rejuvenation. Methods Fractional CO2 and Nd: YAG laser irradiated mice back removed-hair skin one times every ten days, totally four times. After irradiation at the time of two week, one month and three month respectively, the skin tissues in expo-sure area and control area were choosen for immunohistochemical detection of MMP-1 and TIMP-1 ; their dy- namic changes were analyzed. Results Fractional CO2 laser and Nd: YAG laser promoted the production of MMP-1 and TIMP-1 of skins in exposed areas. Both fraction CO2 and Nd: YAG laser, the positive expres-sion MMP-1 and TIMP-1 at two weeks in exposure zone were the most obvious; and fractional CO2 laser was more productive than Nd: YAG laser. Conclusion Fractional CO2 and Nd: YAG laser exposure enhance the expression of MMP-1 and/TIMP-1, and the application of fractional CO2 laser were better than that of t Nd : YAG laser.
