电针水沟穴对全脑缺血昏迷大鼠脑组织谷氨酸转运体-1γ-氨基丁酸转运体-1的影响 被引量:13

摘要 目的观察电针水沟穴对全脑缺血昏迷模型大鼠脑组织内谷氨酸转运体1(EAAT-1)和γ-氨基丁酸转运体-1(GAT-1)表达的影响。方法 80只SD大鼠随机分为假手术组、模型组、针刺组、药物组和针药组;制备全脑缺血性昏迷模型大鼠,针刺组和针药组于造模成功后30min进行首次电针水沟穴治疗,直至处死时间段为止每6h针1次,每次留针时间5min;同时药物组和针药组,于造模成功后30min每只腹腔注射胞磷胆碱钠注射液0.1g,每6h1次。24h后断头取脑,使用计算机图像分析系统测定经免疫组织化学染色处理的大鼠脑组织EAAT-1、GAT-1的平均吸光度(A)值,比较各组之间的差异。结果①各组与模型组和假手术组标本内EAAT-1的A值比较差异均有统计学意义(P<0.05),而模型组和假手术之间差异无统计学意义(P>0.05);针刺组与针药组比较差异无统计学意义(P>0.05),但2组分别与药物组比较差异有统计学意义(P<0.05);②针刺组及针药组与其他各组标本内GAT-1的A值比较差异有统计学意义(P<0.05),模型组与假手术组比较差异有统计学意义(P<0.05),但模型组与药物组之间差异无统计学意义(P>0.05)。结论电针水沟穴可以提高全脑缺血昏迷模型大鼠脑组织内EAAT-1和GAT-1的表达。 Objective The research observed the effect on the expression of excitatory amino acid transporter (EAAT)-1 and γ-amion-butyric acid transporter (GAT)-1 in brain tissue of rats with global cerebral ischemia coma by electro-acupuncturing Shuigou. Methods Eighty SD rats were randomly divided into five groups: control group,model group, acupuncturing group, drug group and both acupuncturing and drug group. Above of all, to prepare the model of whole brain ischemic coma rats; then half an hour later, acupuncturing group and both acu- puncturing and drug group received the treatment of electr-acupuncture Shuigou untill the rats of two groups had been dead,The frequency of acupuncturing was per 6 hours , lasting 5 mins. At the same time, drug group and both acupuncturing and drug group received the treatment of intraperitoneal injection of citicoline sodium injection 0.1 g, Q 6 h. Finally, all 80 rats were executed 24 hours later, the differences were compared by testing the average optical density of EAAT-1 and GAT-1 in brain tissues of rats in all groups. Results ①There were significant differences between model group, control group and other three groups by testing the EAAT-1 values (P〈0.05) the model group and control group had no significant differences (P〉0.05); the difference between acupuncturing group and both acupuncturing drug group was not significant (P〉0.05), but there was significant difference between the two groups and the drug group (P〈0. 05 ); ②Comparing the acupuncture group (or both acupuncture drug group ) and the other groups, there were significant differences by testing the GAT-1 values (P〈0.05) ; the model group was compared with the control group, there were significant differences (P〈0.05), but the difference between the model group and drug group was not significant (P〉0.05). Conclusion Electro-acupuncturing Shuigou increases the expression of EAAT-1 and GAT-1 in whole brain ischemic coma rats.
