不同日龄鸭肝脏中IGF-1及IGF-1R基因表达规律的研究 被引量:5

摘要 为明确鸭肝脏胰岛素样生长因子1(IGF-1)及IGF-1受体(IGF-1R)基因mRNA表达的发育性变化规律,选择0、14、28、42和56日龄高邮鸭和金定鸭各10只(共100只),称测体重后屠宰,采集肝脏组织,采用实时荧光定量PCR法检测IGF-1和IGF-1R基因的发育性变化,并进行品种间比较。结果表明:0~56日龄期间,高邮鸭和金定鸭肝脏IGF-1基因表达量均呈先升后降的趋势,56日龄时高邮鸭肝脏IGF-1基因表达量极显著高于金定鸭;IGF-1R基因表达量基本呈下降趋势,高邮鸭肝脏IGF-1R基因表达量在0日龄时显著高于金定鸭。高邮鸭和金定鸭肝脏IGF-1和IGF-1R基因表达的发育性变化与体重存在不同程度的负相关性。鸭肝脏中IGF-1和IGF-1R基因表达量有特定的发育模式且存在品种差异,IGF-1R基因表达水平的变化不依赖IGF-1基因表达水平。 This study was to find out the developmental patterns of insulin-like growth factor-1 (IGF 1) and IGF-1 re- ceptor (IGF-1R) gene expression in different duck breeds in liver tissue. Real time PCR was applied to investigate and compare the developmental patterns of IGF-1 and IGF-1R gene in liver tissue of duck. 10 of Gaoyou duck and Jinding duck were sampled at the age of 0, 14, 28, 42, 56 day respectively. The results showed that the expression levels of IGF-1 mRNA in liver tissue in Gaoyou duck and Jinding duck increased first and declined thereafter gradually during 0 to 56 day. The expression of IGF-1 gene in Gaoyou duck had very significant difference with that in Jinding duck at the age of 56 day (P〈0. 01). The expression levels of IGF-1R mRNA in liver tissue declined gradually from the age of 0 to 56 day. The expression of IGF-1R gene in Gaoyou duck at the age of 0 day was significantly higher than that in Jinding duck. The developmental patterns of IGF-1 and IGF-1R gene expression in Gaoyou duck and Jinding duck had negative correlation in various degrees with body weight. The IGF-1 and IGF-1R mRNA expression in liver tissue of duck fol- lowed specific developmental patterns respectively. Level of IGF-1R mRNA expression did not depend on that of IGF-1 mRNA expression.
