摘要
目的 :探讨 P38信号通道 (P38MAPK)在脂多糖 (L PS)诱导人单核细胞表达肿瘤坏死因子 (TNFα)中的作用。方法 :采用免疫沉淀法、免疫复合物蛋白激酶测定法、Western blotting检测 P38MAPK在 L PS诱导的人单核细胞产生 TNFα中的激活程度 ,应用逆转录聚合酶链式反应 (RT PCR)和酶联免疫吸附法(EL ISA)观察 P38信号通道抑制剂 SB2 0 35 80对单核细胞 TNFα m RNA和蛋白质表达的影响。结果 :L PS刺激单核细胞引起 P38MAPK激活和 TNFα表达明显增多 ,P38MAPK抑制剂 SB2 0 35 80对单核细胞 TNFα表达有显著抑制作用。结论 :P38MAPK可能是 L PS诱导人单核细胞表达
Objective:To investigate the potential role of P38 mitogenactivated protein kinase( P38MA PK) in inducing proinflammatory cytokine 〔tumor necrosis factorα(TNFα)〕 formation in human monocytes stimulated by lipopolysaccharide(LPS).Methods:Immunoprecipitation,immune complex protein kinase assay and Western blotting were used to detect P38MAPK activity in monocytes stimulated by LPS.Reverse transcription polymerase chain reaction (RTPCR) and enzymelinked immunosorbent assay(ELISA) methods were used to investigate the effect of SB203580 (P38MAPK inhibitor) on TNFα mRNA and protein levels in monocytes.Results:LPS caused activation of P38MAPK at time and dosage dependent manner,and SB203580 inhibited not only activation of P38MAPK but also production of TNFα at time and dosage dependent manner.Conclusions:P38MAPK may play an important role in production of TNFα in human monocytes.
