Quantitative determination of sodium cyclamate in wine samples by UPLC-MS

摘要 目的建立UPLC-MS快速定量检测葡萄酒中甜蜜素的方法。方法采用超高效液相色谱串联三重四极杆质谱仪,色谱条件:Agela Venusil-C18(2.1 mm×50 mm,1.7μm)色谱柱,柱温40℃,流动相为:乙腈=0.01mol/L,乙酸铵=95∶5,流速为0.25 ml/min。质谱条件:ESI源,选择离子监测(SIM)。以定性离子对之间的相对丰度定性,以定量离子对峰面积定量。结果甜蜜素在0.005 0~0.557 4 mg/L范围内呈现良好的线性关系(r=0.999 8);平均加样回收率均在91.14%~100.82%;甜蜜素含量重复性试验RSD<4.779%。结论本方法定性可靠、定量准确,且快速简便,可用于葡萄酒中甜蜜素的检测。 Objective To establish an UPLC - MS method for determination of sodium cyclamate in wine samples. Methods Ultra performance liquid chromatography tandem three quadrupole mass spectrometer was adopted, LC condition: An agla BEH - C18 (2. 1 mm × 50 mm, 1.7μm) column was used with the isocrstic elution of acetonitrile and 0.01 mol/L ammo- nium acetate at a flow rate 0.25 ml/min. MS condition: adopted electrospray ionization(ESI) , and selected ion monitoring, relative intensities of the detected qualitative ion pairs using for qualitative, integrating the signals of the detected quantitative ion pairs using for quantitative. Results The sodium cyclamate had good linearity with correlation coefficient of 0. 999 8,ranging from 0. 005 0 -0. 557 4 mg/L respectively. The average recoveries were 91.14% - 100. 82%. Experiments showed that RSD values of their contents were less than 4. 779%. Conclusion This method is reliable qualitative and quantitative accuracy, simple and rapid, and can be used for wine samples added in sodium cyclamate detection.
