摘要
目的:探讨片仔癀对体外培养的人骨肉瘤U2OS细胞周期的作用及机制。方法:0.8、1.2、1.6mg/ml片仔癀干预U2OS细胞24、48、72h后,MTT检测片仔癀对U2OS细胞的生长抑制作用;片仔癀干预U2OS细胞48h后流式细胞术分析细胞周期的变化;Western blot检测Survivin蛋白的表达。结果:与对照组比较,不同浓度的片仔癀作用24、48、72h后,不同程度的抑制了U2OS细胞增殖,且呈时间和剂量依赖性(P<0.05);片仔癀干预U2OS细胞48h后细胞发生G2/M阻滞,Survivin蛋白表达较对照组明显降低(P<0.05)。结论:片仔癀可抑制骨肉瘤U2OS细胞的生长,诱导U2OS细胞发生G2/M周期阻滞,周期阻滞的发生与Survivin蛋白表达减少有关。
objective:To explore the role of Pien Tze Huang in cell cycle in human osteosarcoma U2OS cells and its mechanism. Meth- ods:U2OS cells were exposed to different concentrations (0.8, 1.2andl.6mg/ml) of Pien Tze Huang for different durations (24, 48and72h), the growth inhibition was measured by MTr assay. The effects of Pien Tze Huang on cell cycle were performed by flow cy- tometry. The Survivin protein expression was detected by Western blot.Results :Compared to the control group,treated with different con- centrations of Pien Tze Huang for different durations significantly reduced cell growth to a varying extent (all P〈0.05) in a dose-time dependent manner.After treatment with Pien Tze Huang for 48h, Analysis of U2OS cell cycle by flow cytometry indicated that Pien Tze Huang could induce cycle arrest in G2/M phase.Western blot analysis indicated Pien Tze Huang down-regulated Survivin protein ex- pression.Conclusion :Pien Tze Huang can inhibit the proliferation of U2OS cells and arrest the cell cycle in G2/M phase, cell cycle ar- rest may be related to survivin orotein exnressiun reduction.
