摘要
目的:探讨缺血预处理(IPC)对抑郁大鼠缺血/再灌注(I/R)心肌细胞凋亡及凋亡基因Fas、Fas-L表达的影响。方法:SD大鼠48只,随机分为非抑郁假手术组(Hsham),抑郁假手术组(Dsham),抑郁缺血/再灌注组(DI/R)和抑郁缺血预处理组(IPC+DI/R);采用原位末端标记法检测心肌细胞凋亡,免疫组化方法和逆转录-聚合酶链反应检测凋亡基因Fas、Fas-L的表达。结果:(1)与Hsham组、Dsham组比较,DI/R组、IPC+DI/R组的心肌细胞凋亡指数显著增加(P<0.001),Hsham与Dsham两组间比较无显著差异;与DI/R组比较,IPC+DI/R组的心肌细胞凋亡指数显著减少(P<0.001);(2)与Hsham组、Dsham组比较,DI/R组、IPC+DI/R组的Fas、Fas-L的mR-NA和蛋白表达均显著增加(P<0.001),Hsham与Dsham两组间比较无显著差异;与DI/R组比较,IPC+DI/R组Fas、Fas-L的mRNA和蛋白表达均显著减少(P<0.001)。结论:IPC能显著减少抑郁大鼠的I/R心肌细胞凋亡,其机制可能与下调Fas、Fas-L基因的表达有关。
Objective: To explore the effects of ischemic preconditioning (IPC) on myocardial apoptosis and the expression of Fas, Fas-L in depression rats that underwent myocardial ischemia/reperfu- sion (I/R). Methods: Forty-eight SD rats were randomly divided into four groups with 12 in each group: non-depression and sham operation group (Hsham), depression and sham operation group (Dsham), depression and myocardial I/R group (Dr/a ), depression, IPC and I/R group (IPC + Dr/a). The myocardial apoptosis was detected by in situ end labeling (TUNEL) method, and the expression of Fas and Fas-L were determined with immunohistochemistry and reverse transcription polymerase chain reaction (RT-PCR). Results: (1)The myocardial apoptosis indexes in group Dr/R and group IPC + Dt/R were significantly higher than those in groups Dsham and Hsham (P 〈 0.001 ). No significant difference was found between group Hsham and group Dsham, and the myocardial apoptosis indexes in group IPC + DI/R were significantly lower than those in group Dt/a (P 〈 0.001 ). (2) The expression of Fas, Fas-L mRNA and protein in group DI/R and IPC + DI/R were significantly higher than those in group Hsham and group Dsham(P 〈0.001 ). No significant difference was observed between group Hsham and group Dsham . The expression of Fas and Fas-L mRNA and protein in group IPC +DI/R were significantly lower than those in group DI/ R ( P 〈 0.001 ). Conclusions: IPC can decrease myocardial apoptosis in depression rats underwent I/R, and the mechanisms might be associated with the expression of down-regulating gene of Fas and Fas-L.
