摘要
羟基磷灰石、壳聚糖和聚乳酸通过原位生成法和溶液共混法制备三元纳米复合支架材料,将293T细胞接种于该支架材料。通过倒置荧光显微镜、扫描电镜观察材料的结构及细胞在该支架材料中黏附、生长、增殖等情况;MTT法检测细胞增殖;以及体外物理性能检测。体外实验证实该支架材料具有良好生物相容性、丰富的孔隙率、良好的力学性能等优点,对进一步开发骨组织工程支架材料应用与临床治疗具有指导意义。
Hydroxyapatite/ehitosan/poly (L-lactic acid) ternary nanocomposites scaffold was prepared by in situ method and solution blending method. In vitro 293T ceils were encapsulated into the scaffold and cultivated in three-dimensional envirnment to observe the biocompatibility of the scaffolds including cell viability, adhesion, proliferation by inverted microscope and SEM. The cell proliferation was tested by MTT method and physical performance was tested by physical method. Live/dead staining with calcein acetoxymethyl ester/Ethidium homodimer-1 ( Calcein-AM/EthD-1 ) was performed in order to determine cell viability on the scaffold. The results showed that hydroxyapatite/chitosan/poly ternary nano biocomposites scaffold display high mechanical property, hydrophilicity and good biocompatibility, suggesting the potential of this composites for applications in bone tissue engineering.
