摘要
目的:通过体外实验的方法探讨苏木精及其提取的4种主要成分对小鼠免疫功能的影响,从而筛选出苏木的免疫活性成分。方法:将苏木精及其分离提取的成分B、C、D、E分为5个实验组,每组样品设4个浓度梯度,分别为0.1mg、1mg、10mg、100mg,同时设立空白对照组及刀豆A单纯刺激对照组。以有丝分裂原伴刀豆蛋白A(ConA)活化小鼠脾脏T淋巴细胞,通过MTT法检测吸光度值和ELISA法测定各受试品培养液中sIL-2R的浓度,检测苏木精及分离提取成分对淋巴细胞增殖反应的影响。结果:苏木精组、成分B、成分C、成分D随药物浓度的增加对刀豆A诱导的T淋巴细胞增殖有明显的影响(P<0.05),其对T淋巴细胞增殖呈现抑制增强。其中成分B、D、E与刀豆A单纯刺激组比较,sIL-2R浓度呈现不同程度的降低,苏木精10mg组及100mg组较刀豆A单纯刺激组sIL-2R浓度降低。结论:苏木精的免疫活性成分包含B、C、D、E;苏木精及其成分B、C、D、E对体外T淋巴细胞增殖有不同程度的抑制作用,并具有剂量相关性。
Objective:To investigate the immunosuppressive function of four components extracted from Hematoxylin L in vitro.Methods:Hematoxylin and its component A,B,C,D and E were divided into five groups according to the random principle,each group of samples of four concentration gradients,respectively 0.1mg,1mg,10mg,100mg,and the establishment of the control group and Con A stimulated of the control group.OD value by MTT assay and ELISA method was to measure sIL-2R concentration in culture medium.Detect hematoxylin and its extracts on lymphocyte proliferation response.Results: With the concentration gradually increased,Hematoxylin,Component B,C and D showed inhibitory effect on T lymphocyte proliferation(P<0.05).Compared with Con A simple stimulation group,sIL-2R concentration of Component B,D and E showed varied degrees of reduction,and Hematoxylin 10mg group and 100mg group reduced,too.Conclusion:Immunosuppressive activity ingredient of Hematoxylin contains B,C,D,E.Hematoxylin and its component B,C,D and E inhibit the proliferation of T lymphocytes and they have a correlation with the dosage.
