阿魏酸钠对大鼠佐剂性关节炎血清中细胞因子IL-15、IL-23、TGF-β的影响 被引量:1

摘要 目的:探讨阿魏酸钠对佐剂性关节炎大鼠血清中TNF-α、IL-10、IL-17水平的影响,来研究阿魏酸钠的干预效果。方法:将清洁级雌性大鼠45只随机分为3组:正常对照组给予以0.9%氯化钠注射液0.1mL于大鼠足跖部,模型组和药物治疗组分别给予0.1%完全弗氏佐剂0.1mL注射大鼠足跖部,在6、12d后予以加强注射免疫。药物治疗组在造模2周后予腹腔注射阿魏酸钠,以50mg/kg每日一次,连续12d;正常对照组与模型组分别给予同等量的0.9%氯化钠注射液。于造模32d后断头处死大鼠,采集血清,酶联免疫吸附法(ELISA)测IL-15、IL-23、TGF-β浓度。结果:模型组与正常组比较,血清IL-15、IL-23水平升高,TGF-β水平降低,治疗组与模型组比较血清IL-15、IL-23表达水平明显降低,TGF-β表达水平上调。结论:阿魏酸钠可降低大鼠血清中IL-15、IL-23的水平,升高TGF-β的水平,对佐剂性关节炎有治疗具有一定的作用。 Objective. To study the effect of sodium ferulate on IL-15、IL-23、TGF-β levels in the serum of adjuvant arthritis rats, and to explore the intervention effect of sodium ferulate. Methods: 45 female Wistar rats were divided into three groups, control group received 0.1 ml doses of 0.9 % sodium chloride injec- tion in the rat paw, while model group and treatment group received 0. 1 ml doses of 0.10% Freund's complete adjuvant injection in the rat paw. After 6 and 12 days, immunity injections were strengthened. 14 days after modeling, treatment group received 50mg/kg doses of sodium ferulate through intraperitoneal injection once a day for 12 days;control group and model group were given equal amount of 0. 9 % sodium chloride injection. 32 days after modeling, rats were killed, and serum was collected, and then the concentrations of IL-15, IL- 23, and TGF-β were measured through enzyme--linked immunosorbent assay (ELISA). Results. IL-15. IL -23 levels were higher and TGF-β levels were lower in adjuvant arthritis rats than those in normal control rats. The expression levels of IL-15,IL-23 in the Sodium ferulate in treated group were significantly lower than those in adjuvant arthritis rats, but the expression levels of TGF-β were increased. Conclusion. Sodium ferulate may reduce IL-15, IL-23 levels in serum, but elevated levels of TGF-β therefore, it has certain effect on adjuvant arthritis treatment.
