摘要
目的 研究幽门螺杆菌 (Hp)全菌抗原的免疫原性及Anti Hp IgY的预防和治疗作用。方法 用超声Hp波粉碎的Hp抗原免疫产蛋母鸡 ,通过IHA和ELISA间接法检测母鸡的免疫应答。用SDS PAGE分析Hp全菌抗原 ,并用抗Hp IgY做免疫印迹。结果 母鸡免疫成功率为 10 0 %。初免 45d后 ,抗Hp IgYIHA效价为 1∶10 2 4,ELISA效价超过 1∶45 0 0 0。Hp抗原的SDS PAGE显示有 2 6条蛋白染色带 ,其中主带有 10条 ,免疫印迹表明Hp的超声粉碎物特异性抗原蛋白带相对分子质量为 70 0 0 0、6 6 0 0 0、42 0 0 0、3430 0、2 95 0 0、2 70 0、185 0 0。结论 本研究为工业化制备抗Hp
Objective To study on the immunogenicity of whole bacterial antigen of Helicobacter pylori and the clinical application of anti Hp IgY .Methods Laying hens were immunized with ultrasonic Hp antigen, and the immune response of them were detected by IHA and indirect ELISA. The whole bacterial antigen was analyzed by SDS PAGE and Western blot with anti Hp IgY. Results The immune response rate of the hens was 100%. Specific anti Hp antibody could be detected out in the yolk laid by the hens 8d after immunization. After a booster, the antibody titers of the hens increased rapidly. At the 45th day after primary immunization, the IHA and ELISA titers reached 1:1024 and above 1:45000 respectively. Twenty six including 10 main bands were observed on the SDS PAGE profile of Hp antigen, and Western blot profile showed the bands of supersonic Hp antigen with molecular weights of 70000, 66000, 42000, 343000, 29500, 27000 and 18500 respectively. Conclusion The study laid a foundation of industrial production of anti Hp IgY.
