乳香活性提取物对成纤维细胞生物学特性的影响

Effects of Frankincense extract on biological behavior of dermal fibroblasts

摘要 目的观察乳香活性提取物对人皮肤成纤维细胞(FB)生物学特性的影响,探讨其促进创面愈合的机制。方法分离培养人正常FB,按照随机数字表法分为乳香活性提取物不同浓度组(5×10-4、5×10-3、5×10-2、5×10-1g/L),含0.25%新生小牛血清的DMEM培养液为对照组。采用噻唑蓝(MTT)比色法检测不同浓度乳香活性提取物对体外培养的FB增殖作用的影响。通过流式细胞仪、实时定量聚合酶链反应(Real-time PCR)分别检测乳香活性提取物最适浓度培养条件下FB的细胞周期变化以及1、111型胶原mRNA的表达。结果乳香活性提取物在5×10-3、5×10-2g/L浓度范围,可显著促进FB增殖,测得吸光度(4)值分别为0.312 ±0. 035,0. 366 ±0.051,与对照组(0.257 ±0.029)比较差异有统计学意义(f值分别为2.67、3. 89,P 〈0. 01),其中在5 × 10-2 g/L浓度时测得4值最高,促增殖作用最为显著,为乳香活性提取物促进FB增殖的最适浓度。此浓度条件下乳香活性提取物可明显促进FB通过G/S及S/G2期限制点,S期(18. 30±3.44)及G2M(9. 60 ± 1。26)期细胞与对照组(10.60 ±1. 72、5. 30 ±0.68)比较明显增多,G0/G1期细胞(71.80 ± 3. 92)与对照组(83. 60 土6.85)比较明显减少(f值分别为:14. 44、8. 97、3. 52,p〈0.05)。5 ×10-2 g/L乳香活性提取物组与对照组比较I、III型胶原mRNA表达明显上调分别为2.416 ± 0. 247比1.000 ±0.081和4. 681 ±0.126比1.000± 0.072,差异有统计学意义(i=4.87、15.31,P〈0.01)。结论乳香活性提取物能显著促进FB增殖,加快FB周期进程,同时可上调I、DI型胶原mRNA的表达,可能与乳香促进创面愈合的机制有关。 Objective To observe the effects of Frankincense extracts on the biological character-istics of human dermal fibroblasts ( FBs) in vitro and explore the possible therapeutic mechanism for woundhealing. Methods The optimal concentration of Frankincense extracts was identified by analysing prolifera-tion activity of human normal FBs that treated with different concentration of Frankincense extracts throughthiazole blue (MTT) colorimetric assay. Cell cycle, collagen I and collagen IH mRNA levels in FBs treatedwith the optimal concentration of Frankincense extracts were detected by using flow cytometry ( FCM) andreal-time polymerase chain reaction (PCR). Results At concentrations of 5 x 10-3 g/L to 5 × 10 -2 g/L, theFrankincense extracts significantly enhanced the proliferation of FBs. The most significant concentration was5 × 10-2 g/L(i =3. 89,P 〈0. 01) ,at which an increased percentage of Gj to S and S to G2 phase cells as wellas an increased level of collagen I and collagen III mRNA were obtained compared to the control group. Con-clusion Frankincense extracts can notably promote the proliferation of FB and accelerating the cell cycle ofFB as well as up-regulating the expression of collagen I and collagen III,which may enhance the process ofwound healing.
