摘要
研究Ca2 +与非细胞体系细胞核重建的关系以及Ca2 +的作用机制 ,结果显示 :Ca2 +对非细胞体系中核装配有抑制作用 ,随着Ca2 +浓度的增加 ,抑制核装配作用增强 ,在Ca2 +作用的早期 (作用 1 0分钟后 )加入EGTA ,可以部分地逆转Ca2 +的抑制作用 ,Ca2 +作用 1小时后再加入EGTA ,不再有逆转作用。对Ca2 +作用机制的研究结果显示 ,不是由于ATP和GTP被沉淀而导致细胞核组装被抑制 ,Ca2 +不是通过激活蛋白酶或激活核酸酶起作用 ,钙调蛋白也未参与细胞核组装的调控 ,Ca2 +并不影响核膜前体膜泡与染色质的结合 。
The relationship between Ca 2+ and nuclear reconstitution in a Ca 2+ free system and the mechanism of Ca 2+ action were studied. The results show that Ca 2+ blocks the cellfree nuclear reconstitution. With the increase of Ca 2+ concentration, the percentage of decondensed chromatin decreases. EGTA is added to stop the action of Ca 2+ after certain times. If EGTA is added 10 min after incubation, nuclear vesicles can attach to the surface of the sperm and form flattened bilayered membrane along its periphery. If EGTA is added 1 hr after incubation, there are only vesicles accumulated along its periphery. No bilayered membrane forms. It seems that this is not because Ca 2+ precipitates ATP and GTP or Ca 2+ activates proteinase or nuclease. CaM is not involved the regulation of nuclear vesicle fusion. Ca 2+ has little effect on the binding process, but blocks the fusion of nuclear vesicles.
