摘要
运用平板筛选法从 2 0株不同来源的菌株中得到一株产组成型漆酶的担子菌 (Basi diomycetesp .)。它在自制DC培养基上产酶能力达到 0 58IU/mL ;振荡培养的产酶效果约是静置培养的 4倍。漆酶的产生与生物量在培养 8d内呈现平行关系 ,但随后酶量并不随之而继续上升。 4g/L的酪氨酸和胰蛋白胨有利于酶的产生 ,分别可使酶活力最高增加到2 49IU/mL和 2 0IU/mL。某些化学试剂对酶的产量尚未达到诱导效果 ,但丁香醛连氮和邻联甲苯胺对酶产量有一定的促进作用。 40℃下刺激 1h后 ,可使酶活力提高 1 3倍。
A constitutive laccase producing fungus was gained from 20 strains of different sources with plate method. Laccase activity reached 0.58IU/mL when the strain grew on DC medium with shaking cultivation, which was superior to stationary method by 4 times. The enzyme production paralleled with biomass in early 8 days, subsequently activities declined slightly though its biomass continued upward. Tyrosine and tryptone at the concentration of 4g/L improved the enzyme production, reaching 2.49 and 2.0IU/mL respectively. Some tested chemical agents seemed not to contribute to the increase of the enzyme production effectively except for syringaldazine and o tolidine. Heatshock at 40℃ for 1h made 1.3 times more enzyme expression.
