祛痰通络汤对早期糖尿病肾病大鼠足细胞podocin表达的影响 被引量:5

Effect of Qutan Tongluo Decoction on Expression of Podocin in Podocytes of Early Diabetic Nephropathy Rats

摘要 目的:观察祛痰通络汤(QTTLD)对糖尿病肾病(DN)大鼠足细胞裂孔隔膜蛋白分子(podocin)表达的影响,探讨其防治DN的作用机制。方法:制作大鼠糖尿病肾病模型,分为6组:正常对照组、模型对照组、贝那普利治疗组、祛痰通络汤低剂量组、祛痰通络汤中剂量组、祛痰通络汤高剂量组,各组分别干预8周。8周后观察各组24 h尿蛋白定量,观察肾脏病理学变化,并采用免疫组化观察肾组织podocin蛋白表达的变化。结果:经过祛痰通络汤治疗后,24 h尿蛋白定量下降(P<0.05);病理改变减轻;podocin蛋白表达增加(P<0.05)。结论:祛痰通络汤对DN大鼠具有一定的肾脏保护作用,其机制可能与恢复肾组织足细胞podocin表达有关。 Objective :To observe the effects of Qutan Tongluo recipe (QTYLD)on the expression of podocin in podo- cytes of diabetic nephropathy (DN) rats. Methods : The rats models of diabetic nephropathy were established by injecting with streptozotocin intraperi - toneally, Wistar rats were randomly divided into 6 groups : the normal control group, the mod- el control group, Benazepril Hydrochloridec group, Qutan Tongluo Decoction low dose group, Qutan Tongluo Decoction me- dian dose group, Qutan Tongluo Decoction high dose group. All the rats were gavaged once daily for 8 weeks. 24 h urinary protein were determined, the protein expression of nephrin was detected by immunohistochemistry, renal tissues were also analyzed by pathological examination. Results : After QTYLD intervention, the levels of 24 h total urine protein were de- creased significantly than those in STZ group(P 〈 0.05). The renal pathological status in QTTLD group was significantly improved and the protein expression levels of podocin were also markedly increased ( P 〈 0.05 ). Conclusion : The kidney injury in the rats with diabetic nephropathy was ameliorated by QTYLD, which may be related to restoration of the protein expression of podocin.
