摘要
目的:探讨高效液相色谱法和紫外-分光光度法检测茴三硫片含量的有效性。方法:高效液相色谱法设定色谱柱为Kromasil+C1 8(250mm×4.6mm),流动相为甲醇-水80∶20,设置检测波长为432nm,分光光度法以432nm处以外标法测定。结果:高效液相色谱法显示茴三硫样品色谱中辅料无干扰,浓度在33.0~252.5μg/mL范围内与峰面积线性关系良好(r=0.998),加样回收试验平均回收率为99.89%,RSD为0.43%;分光光度法显示茴三硫浓度在4.106~20.524μg/mL范围内与吸光度呈良好线性关系(r=0.999),加样回收试验平均回收率为99.87%,RSD为0.46%。两种方法测量3个批次样品茴三硫含量差异无明显差别。结论:高效液相色谱法和紫外-可见分光光度法测定茴三硫含量的准确性均较高,均可用于茴三硫片的质量控制。
Objective, To investigate the validity of high performance liquid chromatography (HPLC) and ultraviolet-visible spectrophotometry for detection of anetholtrithione piece content. Methods. Column for HPLC was set as Kromasil -{- C18 (250 mm )〈 4.6 ram), mobile phase of methanol-water 80 : 20,detection wavelength was 432 nm, spectrophotometry was set at 432 nm external standard meth- od. Results, HPLC accessories concentration was 33. 0-252. 5 μg/mL and showed no interference, the anetholtrithione sample chromatogram peak area had a good linear relationship ( r=0. 998 ) . Average re- covery was 99.89%, RSD was 0.43% spectrophotometry showed that with concentration 4. 106-20. 524 μg/ml, the anetholtrithione had good linear relationship ( r=. 999) and the absorbance Recovery test showed average anetholtrithione photometry are anetholtrithione recovery rate was 99.87%, RSD was 0.46% . No significant difference in the content of was observed among the three samples. Gonclusion: Both HPLC and UV-visible spectro- accurate in anetholtrithione content detection, can be used for quality control of the piece.
