摘要
目的和方法 :为探讨血管活性肠肽 (VIP)对嗜酸性粒细胞功能的调节 ,采用密度梯度离心法分离健康志愿者外周血嗜酸性粒细胞 (Eos) ,培养人脐带静脉血管内皮细胞 (HUVEC) ,然后分别与VIP和IL 1β共育 ,再以血小板激活因子 (PAF)活化Eos,用嗜酸性过氧化物酶 (EPO)活性评估Eos的过氧化反应和脱颗粒反应 ,以白细胞单层粘附试验测定Eos和EC之间的粘附作用。结果 :VIP能抑制PAF致Eos的EPO活性增加效应 ,抑制Eos脱颗粒反应 ,对PAF活化的Eos粘附经IL 1β刺激的EC也有明显的抑制作用。 结论 :VIP对PAF所致Eos功能的易化作用有明显的抑制效应 ,这一作用可能对变应性炎症的治疗具有一定的意义。
Aim:To investigate the effect of vasoactive intestinal peptide (VIP) on the function of eosinophils.Methods:Human eosinophils were purified from periperal blood of normal donator using density gradient centrifugation and treated with VIP,and then stimulated by platelet activating factor(PAF).The cultured human umbilical vein endothelial cells(HUVEC) were stimulated by IL-1β(10u/ml).Eosinophil peroxidase (EPO) assay was used as an index of oxidative burst and degranulation of eosinophil.Eos adhesion was determined by leukocyte-endothelial cell monolayer adhesion asay.Results:VIP could inhibit PAF-induced increase of the EPO activity,and degranulation of eosinophil,VIP also significantly inhibited adhesion of the PAF-activated eosionophils to the IL-1β-stimulated endothelial cells.Conclusion:VIP could attenuate PAF activity that increase the oxidative burst and degranulation of eosinophil,and the adhesion of eosinophil to the EC,which may be of significance in treatment of allergic inflammation.
