摘要
目的 :研究大鼠中度侧位液压冲击脑损伤时脑干c fosmRNA及其表达产物Fos变化规律。方法 :雄性SD大鼠 ,随机分为正常对照组、手术对照组和损伤组。损伤组动物均给以 0 .2MPa液压冲击脑损伤 ,按冲击后处死时间不同再分为 5min、15min、30min、1h、2h、4h、8h和 12h组。应用免疫组织化学和原位杂交方法观察c fos在脑干的表达。结果 :脑冲击后 15min~ 12h ,Fos阳性细胞数逐渐增多 ,12h达高峰。冲击后 5min ,c fosmRNA表达开始增强 ,1~ 2h达高峰。结论 :侧位液压冲击脑损伤后c fos在脑干表达迅速增强 ,持续时间较长。
Aim:To study the expression of c-fos in brain stem following moderate lateral fluid percussion brain injury in rats,and to observe the temporal patterns of its expressions following percussion.Methods:Male Sprague-Dawley rats were divided into normal control,sham operation control and injury groups. The rats of injury groups subjected to moderate lateral fluid percussion injury(0.2 MPa),and then were subdivided into 5 min,15 min,30 min,1 h,2 h,4 h,8 h,and 12 h groups according to the time elapsed after injury.The expression of c-fos was studied by immunohistochemistry and in situ hybridization.Results:After percussion for 15 min,Fos positive neurons increased in brain stem progressively,and peaked at 12 h.At 5 min after percussion,the induction of c-fos mRNA was increased,and remained elevated up to 1~2 h after brain injury. Conclusion:The induction and expression of the c-fos in brain stem after fluid percussion brain injury were increased rapidly and lasted for a long time.
