摘要
我们曾对转基因动物的制作方法、转基因动物研究的应用、转基因的表达特征及提高转基因表达的策略等作过专题综述[1] 。而这之后在转基因动物的研究方面又获得了许多新进展 ,其中转线粒体动物的问世拓宽了转基因动物的研究内容。在制作转基因动物的方法上 ,1998~ 1999年世界上先后报道了三种新方法 :即由Schniek等报道的通过体细胞核移植技术实现转基因 ,由AnthonyW .S .C等报道的通过用逆转录病毒载体感染MII期的卵母细胞实现转基因和由AnthonyC .F .P等报道的通过精子头与待转基因片段一并显微注射卵母细胞的方法实现转基因。在转基因动物的应用上Bernard等将大鼠肠乳糖酶 (根皮苷水解酶 )基因转入小鼠基因组 ,从而使转基因小鼠乳汁中乳糖的含量减少了 5 0 %~ 85 %。此外 ,对转基因表达水平低也有了新的解释。
Some ways of animal transgenic research, such as methodology of animal transgenic makings, application of transgenic research, expression characteristics of transgenes in the genome of transgeic animals and the strategies to improve the expression of transgenes were reviewed by us. After that some new progress had been made in the following years, including the invention of mitochondrial transfer techniques, which brodened the fields of animal transgenic research. As far as the methods for animal transgenic making were concerned, three new methods for animal transgemic making were reported during 1998 and 1999, including gene transfer by nuclear transfer of animal somatic cells, reverse-transcribed gene transfer and intracytoplasmic sperm injection. When talking about the application of animal transgenic research, rat intestinal lactase-phlorizin hydrolase cDNA was integrated into the mouse genome, and lactose in the milk of transgenic mice was reduced by 50% to 85%. There were also some new reports on the explanation of low level expression of transgenes in the genome of their hosts.
