吲哚-2,3-二酮对ox-LDL所致心肌微血管内皮细胞损伤的影响

摘要 目的观察吲哚-2,3-二酮对心肌微血管内皮细胞的保护作用。方法建立大鼠乳鼠心肌微血管内皮细胞(rCMEC)的组织贴块培养法,并对其进行形态学和免疫细胞化学染色鉴定。用氧化低密度脂蛋白(ox-LDL)诱导rCMEC损伤建立损伤模型,随后用吲哚-2,3-二酮作用受损伤细胞,观察其对rCMEC的保护作用。结果倒置显微镜下可见细胞呈梭形并呈铺路石状生长,部分区域可见管腔样结构,CD31相关抗原免疫细胞化学染色鉴定阳性。0.1 g.L-1的ox-LDL对细胞生长有明显的抑制作用,可以作为心肌微血管内皮细胞损伤模型的造模浓度。吲哚-2,3-二酮能减轻ox-LDL对细胞的损伤(P<0.05),并有剂量依赖关系。结论吲哚-2,3-二酮对ox-LDL所致心肌微血管内皮细胞损伤有保护作用。 Objective: To observe the protection effect of indol-2,3-dione on cardiac mlcrovascular endothelial cells. Methods: We established the primary culture method of rat cardiac micmvascular endothelial cell ( rCMEC), and identified the cells with morphology and immune cell chemical dyeing. Then ox-LDL was used to induce the damage and damage mod- el was established. Finally, the protection effect of indol-2,3-dione on the rCMEC was observed. Results: Under the in- vertod microscope, the cells were presented in fuslform shape and grew like the paving stone, and at the same time some re- 8ion had tube-like structure. It was positive for the immunocytochemical staining of factor CD31-related antigen, ox-LDL ( 0. 1 g·L^-1 )inhibited the growth of ceils, and could be used as the concentration of the model. Indol-2,3-dione reduced the damage of ox-LDL in a dose-dependent manner (P 〈0.05 ). Conelusion:Indol-2,3-dione can protect the rCMEC damaged by ox-LDL.
