摘要
常规方法制备原生质体扫描电镜样品时容易造成原生质体变形和破裂,影响其电镜观测结果。本文工作中,通过选用适宜的固定剂对原生质体进行快速固定,然后将原生质体悬浮液滴在经过硅烷化处理而引入了胺基的玻片上,直接在玻片上进行原生质体洗涤、脱水、置换、临界点干燥、喷镀处理,避免了在制样过程中进行离心操作。
Conventional prepartive method is not quite satisfactory with protoplast for SEM observation.In conventional preparation process,protoplast is readily broken or deformed when it is centrifugalized.In order to improve the quality of protoplast observation with SEM,a new preparative method which is free from centrifugalization is introduced in this paper.With this method,protoplast is fixed with a suitable fixing agent.Then,a drop of the suspension solution of the fixed protoplast is put on a thin slice of glass.Which is pretreated with NH\-2CH\-2CH\-2CH\-2Si(OC\-2H\-5)\-3 and has an aminated layer on its surface.Thus,the protoplasts are absorbed and fixed on the glass.Then,the washing treatment,dehydrolysis and replacing treatment of the protoplasts could be proceeded directly on the glass followed by coating with gold.Centrifugalizing operation is avoided during the whole preparation process of protoplast for SEM observation and the protoplasts remian intact.
