摘要 目的探讨一种建立不完全性肠梗阻模型的新方法,以寻求一种稳定可靠的动物模型。方法 SD大鼠随机分为正常组(n=24)、丝线组(n=30)和套环组(n=30)。分别用丝线和套环造成回肠末端梗阻,造模后观测大鼠体重和排便情况,分别于造模后12h、24h、48h和72h开腹腔进行肠道大体形态评分,测量梗阻部位以上肠管周径;取肠组织制作常规石蜡切片,HE染色后观察肠粘膜损伤程度。结果与正常组相比两模型组的粪便颗数和干重在12h和72h均显著下降,梗阻大鼠体重减轻、进食量减少,梗阻上段肠周径逐渐增粗;两模型组的肠道大体形态损伤评分值和肠粘膜损伤评分值均随时间而增高且高于正常组,72h时套环组的两类损伤评分值均显著低于丝线组;套环组的存活率高于丝线组。结论两种方法均建立了不完全性肠梗阻模型,套环法对大鼠肠道损伤更小、安全性高,且适合研究早期不完全性肠梗阻。 Objective To explore a new method to establish a rat model of incomplete intestinal obstruction and to provide a stable and reliable model for future research.Methods SD rats were randomly divided into normal group(n=24),silk group(n=30) and ring group(n=30).The terminal ileum was obstructed respectively by silk or ring.After building the model,the body weight and feces of the rats were observed.We opened abdominal cavity to evaluate the macroscopic intestinal damages and measured intestinal circumference of the upper obstruction respectively after operation at 12 h,24 h,48 h and 72 h;and the intestinal tissue into paraffin section was made,HE stained and then evaluated the intestinal mucosal membrane injury by histological examination.Results Compared with the normal group,feces number and dry weight of two obstructed groups significantly decreased at 12 h and 72 h.Body weight and food intake of two obstructed groups reduced.The circumference of the intestinal which located at upper the obstruction site was gradually thickening;Macroscopic intestinal damage scores and intestinal mucosal injury scores of two obstructed groups were higher than those of the control group.Compared with the silk group,the two injury scores of ring group were lower than control group at each time point and significantly decreased at 72 h;livability of ring group was higher than that of the silk group.Conclusion The two methods were able to establish the model of incomplete intestinal obstruction.Method of ring reduces injury to the rats than the injuries of the silk and this method are more suitable to research the pristine of incomplete intestinal obstruction in the future.
