APE在原位肝移植术后大鼠肝脏中的表达及其与肝细胞凋亡的关系

Expression of APE in rat liver and the relationship between its expression and the hepatocyte apoptosis after liver transplantation

摘要 目的观察大鼠原位肝移植术后肝细胞内脱嘌呤/脱嘧啶核酸内切酶(APE)表达与肝细胞凋亡的关系。方法采用改良的双套管法建立无肝动脉血供的大鼠原位肝脏移植模型,分空白对照组、假手术组、移植后存活3、6、9、12、24h组,采用免疫组织化学方法检测移植后各时间点肝细胞APE蛋白的表达,同时用TUNEL法检测移植后各时间点肝细胞凋亡的情况。结果免疫组织化学显示空白对照组及假手术组肝细胞广泛表达APE蛋白,APE阳性细胞在移植组术后3h即开始减少,其后继续递减,具有时间依赖性,24h时最低。TUNEL阳性细胞在移植组术后3h即可见,6、9和12h计数逐渐增多,并于术后12h达最高峰,术后24h组阳性染色细胞计数与12h组相当。结论肝移植术后肝细胞APE蛋白表达随时间逐渐减少,细胞凋亡在同一部位逐渐升高,这种变化在时间上一致,在部位上重叠,提示APE蛋白减少可能是肝移植术后肝细胞凋亡加剧的重要原因。 Objective: To study the relationship between expression of APE and hepatocyte apoptosis after liver transplantation in rat. Methods: Construct the model of rat orthotopic liver transplantation with modified double catheterization cannula, Rat were randomly divided into groups: control group, sham surgery group and 3, 6, 9, 12, 24 hour after liver transplantation group, immunohistochemistry methods were used to explore the expression of APE, the TUNEL enzyme labeling was applied to observe the change regularity of the hepatocyte apoptosis. Results : The expression of APE was extensive in control group and sham surgery group, decreased after transplant and got the lowest at 24h, showing the time dependent effect, the hepatocytc apoptosis began to increase at 3h after transplantion, got the peak level at 12h and was plat at 24h. Conclusions: The APE positive staining cells gradually decreased while the TUNEL positive staining cells gradually increased at the same study part. This result indicated that the down - regulate of APE may be an important reason for apoptosis accumulation.
