摘要
目的本实验建立了冰冻切片技术制作药用植物组织永久切片的实验方法。方法将材料用FAA固定液固定,经过包埋后,利用冰冻切片机在-18℃--20℃温度下切片,切片经乙醇梯度脱水、番红-固绿染色、二甲苯透明后,用加拿大树胶封片即得。结果应用该方法制作的植物组织永久切片,结构完整而清晰。结论冰冻切片技术是制作药用植物组织永久切片简便有效的方法,应用前景广阔。
Objective To establish the methods of making permanent sections of pharmaceutical botany tissues by cryo-sectioning technique. Method The procedure consisted of several steps as follows： The materials were embedded after fixed by FAA fixation liquid, then frozen and sliced by cryostat microtome betweent -18 ~C and-20~C ~ Result Sections were dehydrated in ascending series of ethanol, dyed with safranine and fast green, cleared in xylene and sealed with canada balsam？ Structure of the section was intact and clear. Conclusion Cryo-section technology was a simple and efficient method to make permanent sections of plant tissues, it has broad prospect of application.
