摘要
目的:探讨糖尿病脑血管病变的发病机理及其与中医毒损脑络学说的相互关系,为开展中医药对糖尿病脑血管病的有效防治提供科学依据。方法:实验分正常对照组、糖尿病组、中药糖宁胶囊治疗组和西药糖适平治疗组。以链尿佐菌素诱导糖尿病大鼠并连续观察24周,同时给予中药和西药灌胃治疗,采用激光衍射法测定大鼠外周血中红细胞变形性,运用放免法测定大鼠脑内TNF-α和ET-1含量;以化学发光法测定脑组织超氧化物歧化酶(SOD)活性,以硫代巴比妥酸法测定丙二醛(MDA)水平。结果:与对照组相比,长期糖尿病患鼠脑中YNF-α、ET-1和MDA含量增高,SOD活性降低,同时外周血中ET-1含量增高,红细胞变形性低下,差异非常显著(p<0.01);糖宁胶囊可有效降低糖尿病大鼠脑内TNF-α、ET-1和MDA含量,提高SOD活性和红细胞变形能力,与模型组比有显著差异(p<0.05-p<0.01)。结论:糖尿病脑血管病变的形成与中医毒损脑络的发病学说密切相关,活血解毒法及组方糖宁胶囊可有效延缓糖尿病脑血管病变,其机理与其提高红细胞变形能力,抑制TNF-α、ET-1的过量释放,保护血管内皮,以及清除氧自由基,减轻脂质过氧化损伤有关。
To discuss the correlation of pathogenesis of diabets cerebrovascular disease and the theory of Naoluo damaged by toxin of traditional Chinese medicine and furnish scientific proof in order to develop to prevent and cure diabets cerebrovascular disease effectively. It is divided into normal controlled group, diabets group, Tangning Capsule of Chinese medicine treated group and western medicine named Tangshiping treated group. Take an observation of big mouse caused by streptozotocin continuously and at the same time bring the treatment of traditional Chinese medicine and western medicine perfused stomach. Compared with the controlled group, the amount of TNF-α, ET - 1 and MDA of diabets mouse brain increased, the SOD activity decreased at the same time. The amount of ET - 1 in peripheral blood increased. The deformability of erythrocyte decreased. It has significant difference(p < 0.01). Tangning Capsule can decrease the amount of TNF -α, ET - 1 and MDA of diabets mouse brain effectively and elevate the SOD activity and deformability of erythrocyte compared with model group, it has remarkable difference (p < 0. 05-p < 0. 01). The formation of diabets cerebrovascular disease is correlected with the theory of Naoluo damaged by toxin of traditional Chinese medicine. The method of improving the blood circulation and detoxofing and grouped medicine Tangning Capsule can delay the diabets cerebrovascular disease. Its mechanism is connected with the increasement of deformability of erythrocyte, inhibition of excessive release of TNF-α, ET - 1 and ET - 1, protection of blood endothelium and eliminition of oxgyen free radical and releasement of lipid peroxidized damage.
