摘要 为了研究猪骨桥蛋白基因(Osteopontin,OPN)组织特异性的表达规律,采用实时荧光定量PCR(Real-time quantitative PCR,qRT-PCR)分析方法,以β-actin基因为内参,对猪OPN基因在心、肝、脾、肺、肾、大脑、小脑、垂体、下丘脑和子宫内膜等10种组织中的mRNA表达水平进行了检测.结果表明,OPN基因在子宫内膜的表达量为最高,其次是在小脑,在心、肝、脾和肺也有不同程度的表达,而在肾、下丘脑和垂体的表达量很少,从而提示OPN基因在子宫内膜中可发挥重要作用. In order to study the tissue-specific expression pattern of porcine OPN gene, mRNA expression levels in ten tissues of Landrace ~ Yorkshire pigs were detected by using real-time quantitative PCR(qRT-PCR), and these tissues were heart, liver, spleen, lung, kidney, cerebrum, cerebellum, pituitary, hypothalamus and endometrium. The results showed that the levels of OPN gene expression in endometrium were the highest and that there were different expression levels in cerebellum, heart, liver, spleen, lung, and rarely in the kidney, hypothalamus and pituitary. The tissue-specific expression pattern of porcine OPN gene suggests that OPN may play an important role in the endometrium.
