混合菌总脱氢酶活性抑制法评价水中污染物的生物毒性

BIOTOXICITY ASSESSMENT OF POLLUTANTS IN WATER BY INHIBITION OF TOTAL DEHYDROGENASE ACTIVITY OF MIXED BACTERIA

摘要 目的探索一种新的适用于野外水中污染物生物急性毒性的快速评价方法,为野外实际应用提供依据。方法选择天然水体混合菌为受试生物,以亚甲蓝脱氢酶活性测定法作为污染物毒性评价方法,研究了8种重要无机有毒污染物对混合菌总脱氢酶活性(DHA)的影响。结果低浓度的Hg2+、Cd2+、CN-对DHA的抑制作用随浓度的升高而增强,质量浓度为0.75 mg/L的Hg2+、Cd2+、CN-对DHA的抑制率分别达到94.2%、97.5%、92.6%。Cr6+在较高质量浓度(10 mg/L)时才对DHA表现出较强的抑制(抑制率60.1%),而Pb2+、As3+、Ba2+、F-在质量浓度达到7.5 mg/L时对DHA也没有明显的抑制作用。结论混合菌总脱氢酶活性抑制法适于野外评价某些重金属如Hg2+、Cd2+及CN-等污染物的生物毒性。 Objective To explore a new method which can be used in the field to rapidly assess biotoxicity of pollutants in water.Methods Mixed bacteria extracted from natural water body were used as experimental organism;methylene blue dehydrogenase activity was used as bioassessment indicator.The effect of eight important toxic inorganic pollutants on dehydrogenase activity was observed respectively.Results The inhibition effect of Hg2+,Cd2+,and CN-on total dehydrogenase activity of mixed bacteria was increased as their concentration increased.When the concentration of Hg2+,Cd2+,CN-reached 0.75 mg/L,the inhibition rate was 94.2%,97.5%,and 92.6% respectively.And when the concentration of Cr6+ even reached higher concentration such as 10 mg/L,the inhibition rate was 60.1%.Moreover,when the concentration of Pb2+,As3+,Ba2+,and F-reached 7.5 mg/L,still little inhibition effect was found.Conclusion The method of inhibition of total dehydrogenase activity of mixed bacteria is suitable for field biotoxicity assessment for some heavy metals especially Hg2+,Cd2+,and CN-pollutants in water.
