摘要
目的:探讨抵挡汤早期干预对实验性2型糖尿病大鼠血管病变中大血管的保护作用。方法:高脂饲料喂养大鼠12周时尾静脉注射STZ制备2型糖尿病大鼠模型,抵挡汤早期干预组、抵挡汤中期干预组和抵挡汤晚期干预组分别于8周、13周和16周给予抵挡汤灌胃,辛伐他汀组和罗格列酮组大鼠于13周分别给予辛伐他汀和罗格列酮灌胃,上述各组给药均至实验24周时结束。ELASA法检测大鼠血清IL-4、IL-13水平,实时荧光定量PCR法检测主动脉TNF-αmRNA表达。结果:与正常对照组比较,模型对照组大鼠血清IL-4与IL-13水平降低(P<0.05);与模型对照组比较,抵挡汤早期干预组和辛伐他汀组大鼠血清IL-4显著升高(P<0.05);同时抵挡汤早期干预组、抵挡汤中期干预组和罗格列酮组大鼠血清IL-13显著升高,(P<0.05);与正常对照组比较,模型对照组大鼠主动脉TNF-αmRNA表达显著升高(P<0.05);与模型对照组比较,抵挡汤早期干预组、抵挡汤中期干预组和罗格列酮组大鼠主动脉TNF-αmRNA表达显著降低(P<0.05)。结论:抵挡汤早期干预能显著上调2型糖尿病大鼠血清IL-4、IL-13水平,降低大鼠主动脉TNF-αmRNA表达,从而抑制大血管病变的炎性损伤,延缓糖尿病大血管病变的发展。
Objective:To observe the protective effects of early intervention with Didang Decoction on the macrovascular lesions of the experimental type 2 diabetic rats.Methods:The diabetic rat models were established after being injected streptozotocin(STZ) into the caudal vein and had fed with high fat diet for 12 weeks.The rats in Didang decoction late group,Didang decoction mid-term group and Didang decoction early group were given Didang decoction from the 16th,13th and 8th week respectively to the 24th week.The rats in simvastatin group and rosiglitazone group were given simvastatin and rosiglitazone respectively from the 13th to 24th week respectively.Then detecting the level of IL-4 and IL-13 in the serum using ELASA method and the mRNA expression of TNF-α in the aorta using Real-time PCR method.Results:Compared with normal control group,the level of IL-4vand IL-13vin the serum was lower and the expression of TNF-αmRNA in the aorta was higher in model control group,and P0.05 respectively;compared with model control group,the level of IL-4vin the serum was higher in Didang Decoction early group and simvastatin group(P0.05),and the level of IL-13 in the serum was higher in Didang Decoction early group,Didang Decoction mid-term group and rosiglitazone group(P0.05);compared with normal control group,the expression of TNF-αmRNA in the aorta was higher in model control group(P0.05);compared with model control group,the expression of TNF-αmRNA in the aorta was lower in Didang Decoction Early Group,Didang Decoction mid-term group and rosiglitazone group(P0.05).Conclusion:Giving Didang Decoction early to the experimental type 2 diabetic rats can raise the level of IL-4 and IL-13 in the serum,and can reduce the mRNA expression of TNF-α in the aorta.Thus the inflammatory injury of macrovascular disease is inhibited,and the development of diabetic vascular disease is delayed.
