摘要
选取猪链球菌2型的cps2J基因设计引物和探针,通过PCR扩增Cy3标记的DNA片段与固定于芯片上的探针进行杂交,并将杂交完毕的芯片进行信号扫描分析,根据荧光信号的强度来确定是否存在猪链球菌2型。结果表明,采用浓度为5μmol/L的探针与PCR产物杂交30min即可得到清晰的荧光信号,表明基因芯片检测技术是一种灵敏度高、特异好的检测方法。该方法的建立可以快速有效地对猪链球菌2型做出诊断,具有较好的应用前景。
The cps2J gene of Streptococcus suis serotype 2 was chosen as a target for the design of primers and probes. Amplified DNA fragments labeled by Cy3 were hybridized with the gene chip fixed with specific probes. Then whether the Streptococcus suis serotype 2 was in or not was detected by scanning the signal obtained from the gene chip and the intensity of fluorescent signal. The method that adopt 5 μmol/L of probe to hybridize with PCR product hybrid for 30 min can get clear fluorescent signal. This indicates that this method is a sensitive and specific method for detecting the disease of Streptococcus suis serotype 2.
