鳖甲煎丸及罗格列酮药物血清对大鼠肝星状细胞TβRⅡ和Smad3mRNA表达的影响 被引量:4

摘要 目的:观察鳖甲煎丸和罗格列酮对由TGF-β1刺激的大鼠肝星状细胞(HSC)内TβRⅡ和Smad3mRNA表达的影响,探讨药物抗肝纤维化的作用机制。方法:采用中药血清药理学的方法,在HSC培养体系中添加外源性TGF-β1,运用反转录聚合酶联反应(RT-PCR)法检测不同剂量鳖甲煎丸和罗格列酮药物血清对由TGF-β1刺激的HSC TβRⅡ和Smad3mRNA表达的影响。结果:TGF-β1上调HSC TβRⅡ和Smad3mRNA的表达,鳖甲煎丸和罗格列酮均下调TGF-β1刺激下的HSC TβRⅡ和Smad3mRNA的表达,鳖甲煎丸作用随剂量增加逐渐增强。结论:鳖甲煎丸和罗格列酮可能是通过在不同位点抑制TGF-β1/Smad信号转导达到抗肝纤维化目的;鳖甲煎丸作用呈剂量依赖性。 Objective:to observe the effect of Bie Jia Jian Wan and Rosiglitazone on HSC TβRⅡ and Smad3mRNA expressions induced by TGF-β1,and to explore the functional mechanism of drugs in resisting hepatic fibrosis.Methods: with seropharmacology,ectogenesis TGF-β1 was added in HSC cultivating system,and RT-PCR was taken to detect the effect of Bie Jia Jian Wan and Rosiglitazone serum of different dosages on TβRⅡ and Smad3mRNA expressions.Results: TGF-β1 increased HSC TβRⅡ and Smad3mRNA expressions while Bie Jia Jian Wan and Rosiglitazone decreased the expressions,the action of which would be stronger with the dosage increased.Conclusion: Bie Jia Jian Wan and Rosiglitazone may resist hepatic fibrosis by inhibiting TGF-β1/Smad signal transduction at different locus;Bie Jia Jian Wan is dose dependent.
